Guide to Normal Phase HPLC Columns

What is Normal Phase Chromatography?

Normal phase chromatography is one of the earliest and most established modes of liquid chromatography. Unlike reversed phase chromatography, it uses a polar stationary phase—often bare silica—and a non-polar mobile phase, such as hexane or heptane. This setup makes it ideal for separating polar compounds based on their interactions with the polar stationary phase.

How Do Normal Phase HPLC Columns Work?

In normal phase chromatography, analytes are separated according to their polarity. Polar compounds interact more strongly with the polar stationary phase and are retained longer, while less polar compounds elute earlier. This retention mechanism allows for efficient separation of isomers, positional variants, and compounds with similar hydrophobic profiles. YMC's Normal Phase HPLC Columns use high-purity silica particles to ensure excellent peak shape and reproducibility. Their wide range of particle and pore sizes allows for high flexibility across different application needs.

When to Use Normal Phase Columns

Normal phase columns are particularly useful when reversed phase chromatography fails to provide sufficient resolution. Typical cases include:

  • Separation of isomers or closely related polar compounds

  • Compounds insoluble in water or polar solvents

  • Analysis of lipophilic substances using non-aqueous mobile phases

Applications of Normal Phase Chromatograph

Normal phase chromatography is widely used in:

  • Natural product analysis

  • Lipid profiling

  • Isomer separation

  • Small molecule and API analysis in pharmaceutical development

YMC's silica-based columns for normal phase chromatography provide reliable performance, high reproducibility, and compatibility with a broad range of organic mobile phases.
