Sensitive LC-MS/MS Analysis of Sphingophospholipids
In LC analyses of sphingophospholipids, such as sphingosine-1-phosphate(S1P) and ceramide-1-phosphate (C1P), phosphate groups in these molecules causes can cause significant peak tailing. Therefore, loss of sensitivity and reproducibility can be observed.
Gowda et al. published a sensitive LC-MS/MS application in the “Analytical and Bioanalytical Chemistry” journal. The application nicely shows that by implementing a metal free column hardware any tailing is avoided.

Efficient Peptide Purification
How to obtain an efficient and reliable purification of peptide pharmaceuticals?
In order to achieve commercial success with any API, consideration of the production costs is essential. Whenever a chromatographic step is involved, purity, yield and throughput are the centre of attention. This Peer Review article summarises all the modern purification techniques comprehensively; definitely a helpful resource for your development work.
You are guided from initial process development, through the stages for optimisation and to the final scale up. Using liraglutide as the prime example, selection of a suitable stationary phase, as well as optimal method conditions are addressed.
