High resolution CEX analysis of commercial monoclonal antibodies
Are you working with monoclonal antibodies (MAbs) and want to characterise them?
Cation exchange chromatography (CEX) is perceived to be the gold standard for the charge sensitive characterisation of MAbs. Acidic and basic variants caused by chemical or enzymatic modifications can be separated from the main isoform of the MAb and have to be critically evaluated.
In this application note, four therapeutic MAbs of different species, isotypes and isoelectricpoint (pI) together with their variants were analysed using pH and salt gradient modes. YMC’s non-porous BioPro IEX SF column was used to characterise Natalizumab (Tysabri®), Cetuximab (Erbitux®), Adalimumab (Humira®) and Denosumab (Prolia®; XGEVA®).

This Technical Note describes three different ADC conjugation types and demonstrates how RP is gaining more importance in the analysis of ADCs and their DAR species.
HIC is the standard technique for the analysis of ADCs and for the determination of drug-to-antibody ratio (DAR). The new generation of ADCs can also be analysed via RP which offers advantages such as direct MS coupling and significantly higher resolution.
