New AAV applications:

HPLC analysis of denaturated and intact adeno-associated viruses via RP-MS and IEX

If you are working with adeno-associated viruses (AAV) these new applications might be interesting for you! These applications show the analysis of two denaturated AAVs by RP-MS as well as the analysis of an intact AAV by IEX.

The denaturated AAVs were analysed using a YMC-Accura Triart Bio C4 column with a wide pore of 300 Å. The three capsid proteins VP1, VP2 and VP3 were successfully separated and characterised using LC-MS with good peak shapes.

Also, the analysis of intact AAV by ion exchange chromatography using a BioPro IEX QF column is described. The empty and full capsid were separated while achieving optimum peak shapes.

New Application Note:

RP analysis of RNA markers using bioinert YMC-Accura Triart Bio C4

This new application note describes the analysis of 7 RNA markers (100 – 1000 bases) using the recently introduced bioinert YMC-Accura Triart Bio C4 column at pH 7 and 80 °C. While conventional column hardware leads to peak tailing and loss of recovery when analysing oligonucleotides, the bioinert coated YMC-Accura Triart Bio C4 column shows excellent recoveries and peak shapes for all 7 well resolved markers.

YMC-Accura Triart (U)HPLC columns

Robust bioinert (U)HPLC columns for high accuracy

The recently introduced YMC-Accura Triart (U)HPLC columns allow superior reproducibility and highest accuracy. They are the ideal choice for analysis of:

  • oligonucleotides, nucleotides
  • peptides and proteins
  • metal coordinating compounds.

Make use of the benefits YMC-Accura Triart columns offer

The unique coating is 130-320-fold thicker making it more durable than other similar hardware concepts. A long-term inertness against sensitive substances is ensured as can be seen in figure A (top area is bare steel for comparison).

Other coated columns can lose their inertness over time. This will again lead to adsorption of sensitive compounds on the uncovered metallic surfaces. Peak tailing, loss of recovery and sample carry over are typical results of the delamination of the coating. In Figure B most of the coating is already delaminated (dark spots: remaining coating).

Employing the bioinert YMC-Accura columns solves this – resulting in sharp peaks and high recoveries from the very first injection to the very last!
