This Technical Note based on the publication of Tsalmpouris et al. presents AEX as reliable and efficient alternative to commonly used fluorescence-based assays for determining the encapsulation efficiency (EE) of mRNA in lipid nanoparticles (LNPs).

This workflow not only increases analytical performance but also yields additional structural insights into LNP composition – delivering added value for advanced pharmaceutical analysis.

The use of bioinert-coated YMC Accura BioPro IEX QF columns further prevents non-specific adsorption, resulting in stable peak areas and reliable quantification.

HILIC as suitable alternative to IP-RP

Hydrophilic interaction chromatography (HILIC) provides an orthogonal alternative to established ion pair reversed-phase (IP-RP) methods – particularly in applications involving mass spectrometry detection.

HILIC offers clear advantages when analysing hydrophilic modifications and performing MS-based detection. It is not only able to separate oligonucleotides by length it is also suitable for the separation of different modifications like phosphorothioated or methoxylated oligonucleotides.
